New JAK inhibitor - CTP-543 by Concert Pharmaceuticals
this new drug was mentioned in the Xeljanz thread but I thought that since it's so difficult to keep track of information on the format of this forum it should have it's own thread
Can we try to keep this thread on topic with any information about CTP-543 that anyone happens to come across?

LEXINGTON, Mass.--(BUSINESS WIRE)--Concert Pharmaceuticals, Inc. (NASDAQ: CNCE) today announced its next product candidate, CTP-543, for the treatment of alopecia areata. Alopecia areata is an autoimmune disease characterized by patchy or widespread hair loss, for which there is significant unmet medical need and currently no FDA-approved treatment. CTP-543 is a deuterium-modified analog of ruxolitinib, which selectively inhibits Janus kinases 1 and 2, known as JAK1 and JAK2. Ruxolitinib is commercially available under the name Jakafi® in the United States for the treatment of myelofibrosis and polycythemia vera. Concert expects to initiate Phase 1 clinical evaluation of CTP-543 in the second quarter of 2016 with efficacy studies commencing in 2017.
Ruxolitinib has been used to treat alopecia areata in academic settings, including an investigator-sponsored clinical trial, and has been shown to promote hair growth in individuals with moderate to severe disease. Preclinical studies conducted by the Company demonstrated that CTP-543 retains ruxolitinib’s selective JAK 1/2 inhibition profile and shows improved metabolic stability.

Recently, the U.S. Patent and Trademark Office issued U.S. Patent No. 9,249,149 that claims CTP-543 as a novel composition of matter.

i was started the trial on 8/2018. now am on the extension study until it comes to market. i was semi responsive to xeljanz and have 95% regrowth now. I had aa (which moved to AT) for about 20 years. no side effects. Ophiasis pattern loss.

Did you have decent regrowth? Did you decide not to continue with Xeljanz?

i had pretty decent growth (around 70% from 20%), then it began to fall out again. I was never able to get my insurance to ok an increase in dosage so once it started falling out I decided to just bail and cut it all off until I found the clinical trial. I have seen that some of the other xeljanz users were able to bump up their dosage of xeljanz and pulse with prednisone at that point. Unfortunately, not an option for me.

I'm hoping they start the study nearby sometime soon. With covid 19 they paused the study because the Cleveland Clinic had to focus on other areas. Weirdly enough I'm starting to get hair back on my face (to the point that I'll have to shave) and my scalp. the scalp hairs are the little white ones so maybe they will mature but as of now it's a waiting game. I think the clean keto diet is helping heal my body.

i did the study in indy (3 hours away) and they paused for a month or so but re-upped at end of May with lots of precautions. but this place does only derm clinical trials so no competing trials to prioritize. All of my scalp hairs started as white but matured (felt slow to me). My nurse said i was a slow responder but ophiasis pattern is stubborn anyway I'm hoping OH is good. . I heard they did very well with the sequester so..fingers crossed you'll get started again soon! all the best!

I did not respond to Xeljanz but have just been approved to start Jakafi. Did you have any side effects?

no side effects at all. i did see on another thread that someone in the study had elevated potassium levels..

Hey mtran2000 - I took Xeljanz for a year and did not respond at all - and had no side effects. I have now been on Jakafi for a year and have full regrowth! But I do have side effects - weight gain, high cholesterol, and fatigue. I now take a statin fore the high cholesterol.
